A standard method for reconstituting peptides for intranasal use, with dilution ratios, spray-volume math, sterility handling, and storage limits.
A standardized clinical framework for reconstituting peptides for intranasal delivery. It defines dilution ratios, spray-volume calculations, sterility handling, and stability parameters so that dosing is precise and reproducible across treatment cycles. All calculations assume a calibrated nasal spray pump that delivers about 0.1 mL per actuation.
Intranasal delivery can allow rapid systemic absorption and possible central distribution through the olfactory and trigeminal pathways, so dosing accuracy is critical.
Each spray delivers about 0.1 mL. All microgram calculations below come from this fixed spray volume.
If you use a different spray device, test its calibration before clinical use.
| Peptide strength | Example | Diluent and peptide | Result |
|---|---|---|---|
| 2 mg | Oxytocin | 2 mL solution + 2 mg peptide | About 100 mcg per spray, about 20 sprays |
| 2 mg | Oxytocin | 1 mL solution + 2 mg peptide | About 200 mcg per spray, about 10 sprays |
| 5 mg | 5 mg peptides, or 5 mg/5 mg blends such as Semax/Selank | 5 mL solution + 5 mg peptide | About 100 mcg per spray |
| 10 mg | 10 mg peptides, 10 mg/10 mg blends, or dual 10 mg combinations such as BPC-157 and KPV | 10 mL solution + 10 mg peptide | About 100 mcg per spray, about 100 sprays |
| 10 mg | As above | 5 mL solution + 10 mg peptide | About 200 mcg per spray, about 50 sprays |
| 50 mg | 10 mL solution + 50 mg peptide | About 500 mcg per spray | |
| 100 mg | GHK-Cu | 10 mL solution + 100 mg peptide | About 1 mg per spray |
Use a sterile syringe and needle for every withdrawal. Avoid puncturing the vial repeatedly beyond safe handling limits. Do not use the solution if it looks cloudy or discolored, or if it contains particles.
Store filled nasal spray bottles in the refrigerator (2–8°C). Avoid light exposure and repeated temperature changes.
Discard any reconstituted intranasal peptide solution after 30 days, regardless of how much remains.
Intranasal administration bypasses first-pass hepatic metabolism and can allow rapid systemic distribution. Because absorption happens across a highly vascular mucosal surface, small differences in dilution can change the delivered dose significantly. Standardized reconstitution gives predictable microgram delivery per spray and makes results more consistent across clinical applications.
